Regulation Genes in Plant Anthocyanin Synthesis Pathway

Anthocyanin, which existed widely in higher plants, is a water soluble plant pigment, and closely related to crop quality traits. Its biosynthetic pathway and the mechanism of related regulation proteins interaction was found in recent years with the further study of the mutants of Arabidopsis. The research of anthocyanins in flowers, fruits and seeds is always the hot spot. Recently there are many research reported, its deepening research can provide scientific basis for health care, and help us to use in many fields. Here we give reviews on the study status of plant anthocyanin biosynthetic genes, including plant anthocyanin biosynthetic pathway, the regulation of transcription factors in biosynthetic pathway, and the regulatory genes which have been separated and cloned.

Research Progress of Plant bHLH Transcription Factor Family

The bHLH gene family is one of the biggest transcription factor families in plant. It plays important roles in the regulation of plant development and stress response. Previous studies showed that the functional research of bHLH in animals is very booming, but there is only little information available in plants. In this review, we surveyed the recent research progress of bHLH transcription family in plant stress response, growth and development. This study will offer some suggestions for the further research of bHLH family in the regulation of plant response to environmental stresses.

This review overviewed mainly about the kinds of herbicide resistance genes, their main resource and the application of herbicide resistance genes. Moreover, we also discussed the discovery of the novel herbicide resistance genes, the modification of herbicide resistance gene using genetic and protein engineering, the mechanisms of herbicide resistance and new herbicide resistance transgenic crops breeding.

Abiotic stress is the most important factor that affect plant growth and development, plant stress resistance mechanism is a main area of plant molecular biology. With the development of molecular biology techniques, the research on plant abiotic stress has made great progress, and the molecular mechanism of plant response to stress has been revealed initially. This review discusses the abiotic stress related mechanism and molecular biology of Ammopiptanthus mongolicus under the drought, cold and salty stress, in order to identify more resources and new stress tolerance genes for plant genetic engineering and to provide ideas of plant stress resistance research.

Stay green characteristic that is related to leaf senescence in plants affacts plant growth and biological production. Stay green reseach is main focused on model plants such as Arabidopsis thaliana, but less in maize. It is of great significance on corn yield, quality and disease resistance to do stay green reseach in maize. This paper reviews recent development in stay green research in maize, including concept of stay green and its evaluation indexes, physiological, biochemical and genetic characteristics, and QTL mapping for this important trait. Some problem in study of stay green in maize and its countermeasures are also discussed.

Non-coding RNAs (ncRNAs) have been recognized as important regulators, whose function is as co-ordinators of adaptation progresses in response to environmental changes. In bacteria, ncRNAs play important roles to regulate diverse processes including sugar metabolism, envelope stress response quorum sensing and virulence. The regulation model has been an important “branch” of bacterial regulatory networks. This paper reviewed research progress on ncRNAs in recent years, and described in detail the synthesis of bacterial ncRNAs and regulatory mechanism. Due to the unique structure of ncRNAs and complexity of regulatory network, the function research of ncRNAs has become one of the hot points in recent year.

Influence of Chemical Pretreatment Technology on the Digestibility of Lignocellulose

Ethanol from lignocellulosic materials is the important developing orientation of bioenergy. In order to increase the digestibility of cellulose and hemicellulose, methods related with the removal of lignin from lignocellulose had been widely studied in recent years, which were focused on the pretreatment of biomass with acids, alkalis, oxides, organosolvs, ionic liquids and so on. Chemical technology for removing lignin at modest temperature and pressure will certainly accelerate the industrialization of cellulosic ethanol.

Screen and Expression of the Penicillium sp. FJ2 Polygalacturonase gene in Escherichia coli

Polygalacturonases are a class of pectinase that can hydrolyze pectin, and they are widely used in food, feed, textile and paper industry. A Penicillium sp. FJ2, which produced polygalacturonase, has been screened in this research. In addition, a polygalacturonase gene pgp1 has been cloned using degenerated PCR and TAIL-PCR. Length of the polygalacturonase pgp1 gene and its cDNA are 1 225 bp and 1 104 bp, respectively. The cDNA of pgp1 encodes 367 amino acids and a termination codon. Pgp1 was cloned into pET-30a(+) vector and expressed in Escherichia coli BL21(DE3). The recombinant PGP1 was expressed as inclusion bodies without activity towards polygalacturonic acid. The renaturation of the PGP1 was through with urea resolution and gradient dilution. Polygalacturonase activity of the renatured PGP1 was detected as 12.9 U/mL, and with specific activity of 5.83 U/mg.

Observation of the Fruiting Body Formation of Myxococcus Fulvus by Microscope Technic

Life cycle of Myxococcus fulvus was observed with stereomicroscope (SMC) and scanning electron microscope (SEM) by culture it within eight days. The results revealed the process of vegetative cells moveing thgether to form the fruiting body. SMC observation indicated the swarm of Myxococcus fulvus gathered around to form the fruiting body and the color of them turned into bright orange. SEM observation indicated that a pile of cells moved together toward one diretion and heaped up to build up the huge fruiting body. Also, the vegetation cells turnned into the spherical myxospores.

Chromosome Doubling Technique for Anther Culture of Winter Wheat

In order to increase chromosome doubling frequency for anther culture of winter wheat, the callus, roots of regenerative plants and tillering node were treated respectively with different concentration of colchicine. The result showed that there were 333%～61.5% setting percentage in the treated callus with 0.02‰ and 0.05‰ colchicine soluion. After soaking roots of regenerative plants for 5 hours using 0.02‰ and 0.05‰ colchicine soluion, the average setting percentage reached 375％. The setting percentage were over 50％ in tillering node soaked for 5～10 hours using 0.04‰ colchicine soluion and 1‰dimethylsulfoxide.
