Transformation liquid was obtained from the reaction of crude enyme with maltose as oligomer. The crude enyme was fermented by trehalose synthase producing strain which was selected from soil. The concentration of trehalose in the transformation was determined by HPLC with Sugar-PakTM Ⅰ. Results showed that the method could separate the trehalose and glucose completely, accordance with the result of DNS colorimetric method of 1.5g/100mL, but the HPLC operation process and data processing were simpler and easier to control in trehalose content determination.
