Abstract:

Humanized zebrafish models can accurately simulate human physiological and pathological processes, effectively overcome interspecies research barriers, and have become important research carriers in biomedicine. On this basis, the paper systematically elaborated on the inherent advantages of zebrafish for constructing humanized animal models, and comprehensively summarized the technical principles, characteristics and strengths of three commonly used approaches for establishing humanized zebrafish models, namely clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9(CRISPR/Cas9), Tol2 transposon system, and xenotransplantation. It further highlighted the research progress and latest breakthroughs of humanized zebrafish models in multiple disease areas, including cardiovascular diseases, tumors, neurological disorders and liver diseases. Potential future research prospects were also proposed, including the combined application of diverse models and the integrated innovation of multiple technologies. The review aims to provide technical references for researchs in humanized zebrafish models and further advance studies on pharmaceutial research and development.

Key words: humanized zebrafish models, gene editing, transposon, xenotransplantation
