Evaluation of the Preventive and Reparative Effects of N-PS on Skin Barrier Impairment

Abstract:

This study aimed to investigate the preventive and reparative effects of Nocardia rubra cell wall skeleton polysaccharide components (N-PS) on skin barrier impairment. The antioxidant activity of N-PS was evaluated, together with its effects on phagocytic activity of RAW 264.7 cells, melanin synthesis and tyrosinase activity in B16 mouse melanoma cells, as well as the reparative effect on HaCaT cells injured by H2O2. Within the concentration range of 0.25~4.00 mg·mL-1, N-PS showed strong DPPH free radical scavenging activity, with a maximum scavenging rate of 40.0%. Stimulation with 200 μg·mL-1 N-PS increased the proliferation rate of RAW 264.7 cells by 2.56%, enhanced the phagocytic activity by 26.94%, and elevated the macrophage-mediated innate immune response by 65.88%. Meanwhile, N-PS reduced the proliferation of B16 mouse melanoma cells by 36.89%, inhibited melanin synthesis by 39.05%, and decreased total cellular tyrosinase activity by 38.86%. N-PS exhibited significant protective effects on H2O2-damaged HaCaT cells, and the cell viability increased from 65.62% in the blank control group to 97.79% after pretreatment with 800 μg·mL-1 N-PS. In summary, N-PS showed a maximum DPPH scavenging activity of 40.0% at 0.25~4.00 mg·mL-1, and 200 μg·mL-1 N-PS exerted favorable preventive and reparative effects on skin barrier impairment.
