Identification and Phylogenetic Analysis of Virus Species Infecting Ornamental Jasminum sambae in Fuzhou Region

Abstract:

Jasminum sambac is an important ornamental plant in China and the city flower of Fuzhou. To identify viral pathogens infecting ornamental jasmine in Fuzhou, virus-suspected jasmine samples from parks in Fuzhou were tested using RT-PCR, and the virus gene sequences were subjected to phylogenetic analysis. Results revealed four viruses in 80 tested sample, jasmine virus A (JaVA), jasmine virus C (JaVC), jasmine virus H (JaVH) and jasmine virus T (JaVT), with detection rates of 18.75%, 51.25%, 30.0% and 21.25%, respectively. Mixed infections were observed in 30.0% samples, with nine different combination patterns showing following frequencies: JaVA+JaVC (3.75%), JaVA+JaVH (1.25%), JaVC+JaVH (5.0%), JaVC+JaVT (2.5%), JaVT+JaVH (1.25%), JaVA+JaVC+JaVH (1.25%), JaVA+JaVC+JaVT (1.25%,), JaVC+JaVH+JaVT (8.75%), and JaVA+JaVC+JaVH+JaVT (5.0%). Sequence analysis showed that the Fuzhou isolates from ornamental jasmine share 98.83%~99.48%, 79.64%~95.29%, 85.84%~97.94% and 92.59%~97.85% nucleotide identity with the respective reference sequences in GenBank for JaVA, JaVC, JaVH and JaVT. Phylogenetic analysis indicated that the JaVC isolates obtained in this study were most closely related to the reported isolate TZS (GenBank accession number: ON241322), the JaVH isolates were most closely related to the reported isolate Jiangsu (GenBank accession number: MH231179), and the JaVT isolates were most closely related to the reported isolate YJ (GenBank accession number: MZ962345). The main virus species and their co-infection characteristics of ornamental jasmine in Fuzhou were reported for the first time, which provids baseline data for the prevention and control of jasmine virus disease.

茉莉花(Jasminum sambac)是我国重要的观赏花卉,同时也是福州市市花。为明确福州地区观赏茉莉花易感染的主要病毒种类,利用RT-PCR方法对福州市主要公园采集的疑似感染病毒病的茉莉花样品进行检测,并对病毒基因序列进行系统发育分析。结果表明,福州地区的80份茉莉花疑似病样中共检测出茉莉花A病毒(jasmine virus A,JaVA)、茉莉花C病毒(jasmine virus C,JaVC)、茉莉花H病毒(jasmine virus H,JaVH)和茉莉花T病毒(jasmine virus T,JaVT)4种病毒,检出率分别为18.75%、51.25%、30.0%和21.25%。其中,30.0%的样品存在复合侵染现象,分别为JaVA+JaVC(3.75%)、JaVA+JaVH(1.25%)、JaVC+JaVH(5.0%)、JaVC+JaVT(2.5%)、JaVT+JaVH(1.25%)、JaVA+JaVC+JaVH(1.25%)、JaVA+JaVC+JaVT(1.25%)、JaVC+JaVH+JaVT(8.75%)和JaVA+JaVC+JaVH+JaVT(5.0%)。JaVA、JaVC、JaVH、JaVT福州茉莉花分离物序列与GenBank已报道的各病毒序列一致性分别为98.83%~99.48%、79.64%~95.29%、85.84%~97.94%和92.59%~97.85%。系统发育分析结果显示,获得的JaVC分离物与已报道的TZS分离物(GenBank登录号:ON241322)亲缘关系最近;JaVH分离物与已报道的Jiangsu分离物(GenBank登录号:MH231179)亲缘关系最近;JaVT分离物与已报道的YJ分离物(GenBank登录号:MZ962345)亲缘关系最近。首次报道了福州市观赏茉莉花的主要病毒种类及其复合侵染特征,为茉莉花病毒病的监测与防控提供了理论依据。
