摘要:

嵌合抗原受体T细胞(chimeric antigen receptor T-cell,CAR-T)疗法作为一种革命性的免疫治疗方法,已在血液系统恶性肿瘤治疗中展现出显著的临床效果。然而,传统的体外CAR-T疗法因存在的众多问题,限制了其广泛应用。近年来,直接体内改造T细胞的CAR-T疗法有望克服传统疗法的局限性。综述总结了体内CAR-T疗法的技术原理,及其最新研究进展、临床应用潜力、安全性问题和未来发展方向,旨在为恶性肿瘤临床研究提供全面的参考,并推动这一创新疗法的进一步优化与转化。

Abstract:

Chimeric antigen receptor T-cell (CAR-T) therapy, as a revolutionary immunotherapy method, has demonstrated remarkable clinical efficacy in the treatment of hematological malignancies. However, the traditional in vitro CAR-T therapy is limited in wide application due to numerous problems. In recent years, the research on in vivo CAR-T therapy, which directly modifies T cells in the body, holds the promise of overcoming the limitations of traditional therapies. This article reviewed the technical principles,latest research progress, clinical application potential, safety issues, and future development directions of CAR-T therapy in vivo, aiming to provide comprehensive references for clinical research on malignant tumors and promote the further optimization and transformation of innovative therapy.
