中草药虎杖离体快繁体系的优化

摘要:

为快速扩繁高质量虎杖种苗,优化虎杖离体组织快繁体系,以带腋芽虎杖茎段为外植体,探究了不同消毒时间和不同激素浓度对虎杖茎段增殖诱导和不定芽生根的影响。结果表明,虎杖带腋芽茎段的最佳灭菌方式为75%酒精15 s+(0.1%吐温-20+2%次氯酸钠)震荡清洗12 min,无菌外植体的获得率达到90.89%。诱导茎段腋芽出芽的最适培养基为MS+30 g·L-1蔗糖+7 g·L-1琼脂+1.5 mg·L-1激动素+0.2 mg·L-1萘乙酸,增殖率达到113.3%。不定芽生根培养的最适培养基为1/2 MS+30 g·L-1蔗糖+5.5 g·L-1琼脂+3 mg·L-1绿色植物生长调节剂GGR6号生根率可达70.37%,且3 d左右分化出不定根,15 d左右获得完整植株;生根后组培苗的移栽成活率达到100%。研究结果优化了虎杖茎段组培快繁体系,无菌外植体获得率、增殖率和移栽成活率均超过90%,并且生根时间快,可显著缩短移栽时间,降低繁殖成本。

Abstract:

To rapid propagate high-quality Reynoutria japonica Houtt. seedlings and optimize a rapid propagation system of Reynoutria japonica Houtt. in vitro tissue, the effects of different disinfection times on the stem segments of Reynoutria japonica Houtt. and different hormone concentrations in the culture medium on the proliferation induction and adventitious bud rooting of the stem segments of Reynoutria japonica Houtt. were investigated. The results showed that the best sterilization method for the stem segment with axillary buds of Reynoutria japonica Houtt. was 75% alcohol 15 s+(0.1% Tween-20+2% sodium hypochlorite) for 12 min, and the acquisition rate of sterile explants reached 90.89%. The optimal medium for inducing axillary bud sprouting in stem segments was MS+30 g·L-1 sucrose+7 g·L-1 agar+1.5 mg·L-1 kinetin+0.2 mg·L-1 naphthalene acetic acid, with a proliferation rate of 113.3%. The optimal medium for rooting culture of adventitious buds was 1/2 MS+30 g·L-1 sucrose+5.5 g·L-1 agar+3 mg·L-1 green plant growth regulator (GGR6). The rooting rate could reach 70.37% and adventitious roots could be differentiated after about 3 days, and complete plants could be obtained after about 15 days. The survival rate of transplanted tissue cultured seedlings after rooting reached 100%. This study optimized a rapid culture system for stem segments of Reynoutria japonica Houtt., with a sterile explant acquisition rate, proliferation rate, and transplant survival rate of over 90%. Moreover, the rooting time is fast, which can significantly shorten the transplant time and reduce reproduction costs.
