低成本、可放大的SUMO/pelB融合表达硫酸软骨素酶B的制备工艺

A Low-cost and Scalable Preparation Process for SUMO/pelB Fusion Expression of Chondroitinase B

摘要:

硫酸软骨素酶B(chondroitinase B,ChSase B)是一种钙依赖型裂解酶,能够特异性降解硫酸软骨素B。通过构建pelB/SUMO/ChSase B串联融合表达系统,开发了一种高产量、高纯度、低成本且易于放大的酶制备工艺。通过信号肽pelB引导周质分泌和小分子泛素修饰蛋白(small ubiquitin-like modifier,SUMO)标签增强可溶性表达,在大肠杆菌BL21(DE3)中实现了高效表达。采用国产新型亲和填料结合阴离子交换层析,建立了无重金属污染的纯化流程,最终获得高效液相色谱(high performance liquid chromatography, HPLC)纯度95.4%、比活力374.87 U·mg-1、发酵液产量达到873.14 mg·L-1的冻干酶制剂。工艺放大试验证实,从5 L到30 L规模单位产量提升了35%,冻干粉在-20 ℃储存12个月活性保留率>90%。该工艺具有环境友好性和产业化潜力,为ChSase B的规模化生产提供了可靠的解决方案。

Abstract:

Chondroitinase B (ChSase B) is a calcium-dependent endopeptidase that can specifically degrade chondroitin sulfate B. In this study, a high-yield, high-purity, low-cost, and scalable enzyme preparation process was developed by constructing a pelB/SUMO/ChSase B tandem fusion expression system. Through the guidance of the signal peptide pelB for periplasmic secretion and the small ubiquitin-like modifier (SUMO) tag to enhance soluble expression, efficient expression was achieved in Escherichiacoli BL21(DE3). Using domestic new affinity resins combined with anion exchange chromatography, a purification process without heavy metal contamination was established. Finally, a freeze-dried enzyme preparation with an high performance liquid chromatography purity of 95.4%, specific activity of 374.87 U·mg-1,and fermentation liquid yield of 873.14 mg·L-1was obtained. Process scale-up experiments confirmed that the unit production increased by 35% from 5 L to 30 L, and the activity retention rate of the freeze-dried powder at -20 ℃ for 12 months was >90%. This process is environmentally friendly and has industrialization potential, providing a reliable solution for the large-scale production of ChSase B.
