Abstract: Total microbial DNA was directly extracted from a metamorphic waterborne coating, the clone library of 16S ribosomal RNA genes was amplified using PCR with universal bacterial primer sets. The PCR products were then subcloned into pGEM-T vector, amplified DNA was used for diversity analysis. The total of 96 clones were selected and 8 kinds of microbe which were used for sequencing. Pseudomonas was the most one (75% of total clones), then was Paenibacillus chondroitinus (15% of total clones), Aeromonas caviae and Alcaligenes faecalis accounted for 2% of total clones respectively. 16S rRNA gene library is a good research method for waterborne bacteria community diversity analysis.

摘要： 采用未培养技术直接提取变质涂料微生物总DNA，使用通用引物进行PCR扩增、纯化、连接、克隆和测序，建立16S rRNA基因文 库，分析变质水性涂料细菌的数量与种类。从96个克隆子中共检测到8种不同种属的细菌，假单胞菌属(Pseudomonas)为优势菌 群，占总数的75%，软骨酸芽孢杆菌(Paenibacillus chondroitinus)占总数的15%，此外豚鼠气单胞菌(Aeromonas caviae)和 粪产碱杆菌(Alcaligenes faecalis)分别占总数2%。结果表明16S rRNA基因文库是一种较好的研究水性涂料细菌群落多样性的 方法。
