摘要:

溶瘤病毒(oncolytic viruses, OVs)作为一种新兴的肿瘤免疫治疗工具,通过选择性裂解肿瘤细胞和激活抗肿瘤免疫的双重机制,已在多种实体瘤治疗中展现出潜力。近年来,溶瘤病毒与生物传感器的结合为癌症诊疗提供了新思路:通过基因工程改造,溶瘤病毒可作为生物传感器的动态响应元件,实时监测肿瘤微环境(tumor microenvironment, TME)的分子特征,或作为递送载体将生物传感器靶向肿瘤区域,实现治疗与监测的协同。系统综述了溶瘤病毒的作用机制和主要应用领域,以及作为生物传感器的技术基础和相关的临床研究进展,并展望了未来的发展方向。

Abstract:

Oncolytic viruses (OVs), as an emerging therapeutic tool in tumor immunotherapy, have demonstrated promising potential in treating various solid tumors by selectively lysing tumor cells and activating anti-tumor immunity. In recent years, the integration of oncolytic viruses with biosensors has introduced a novel approach for cancer diagnosis and treatment: through genetic engineering, OVs can serve as dynamic response elements in biosensors to monitor molecular features of the tumor microenvironment (TME) in real time, or act as delivery vectors to target biosensors to tumor sites, thereby achieving synergistic treatment and monitoring. This paper systematically reviewed the mechanisms of oncolytic viruses and their primary applications, the technical foundations of biosensors, and recent clinical advances, while also prospected future directions.
